Absorption and Clearance
Subcutaneous bioavailability is approximately 55%, with peak plasma concentration reached 8–12 hours after dosing. The fatty-acid albumin-binding mechanism slows both absorption and clearance, giving liraglutide its ~13-hour half-life — long enough for once-daily dosing but far shorter than the fatty-diacid modification used in semaglutide, which extends half-life to about 7 days. Steady state is reached within 4–5 days of consistent daily dosing.
