Exendin-4 Origin
Exenatide is a synthetic analog of exendin-4, a peptide first isolated from the venomous saliva of the Gila monster (*Heloderma suspectum*). It shares only 53% sequence homology with human GLP-1 — far less than semaglutide or liraglutide — and this divergence is exactly what makes it naturally resistant to DPP-4 enzymatic degradation, without requiring the fatty-acid modifications used in later-generation GLP-1s.
GLP-1 Receptor Effects
Like other GLP-1 receptor agonists, exenatide enhances glucose-dependent insulin secretion, suppresses inappropriate glucagon release, slows gastric emptying, and promotes satiety. It was the first compound in this class to reach market, establishing the therapeutic template that semaglutide, tirzepatide, and dulaglutide later built on.
